Trypanosoma cruzi causes Chagas’ disease, an important health problem in Central and South America affecting around 10 million people. Allopurinol is  a compound acting in Trypanosoma cruzi as a purine analogue inhibiting the synthesis of mRNA and protein the parasite, but their failure on avoiding Chagas disease progression may be due in part to metabolism process, and to unfavorable physicochemical properties, leading to versatile responses in humans. In the present study, a simple and sensitive method by MLC was optimized and validated for the simultaneous analysis of Allopurinol and their four new derivatives, which are the third family of derivatives synthesized in our research group to overcome Allop’s problems. For the study we using C18 ODS column with UV detection (l= 250nm), at 25ºC, the flow rate was 1.0 mLmin-1, and a micellar solution of 0.10M sodium dodecyl sulphate–7.5 v/v propanol–0.01M NaH2PO4-H3PO4 (pH 3.0) as mobile phase. The analytical separation was performed in less than 10 min, and the parameters studied include linearity (r2> 0.9998), intra- and inter-day precision (RSD, %: 0.21-1.98 and 0.21-1.97, respectively) and robustness. This method was sensitive with limit of detection between 1.7 x 10-10 and 4.0 x 10-10 molmL-1 and limit of quantification was 5.0 x 10-9 molmL-1 for all compounds, with recoveries ranging from 95.53 to 102.86%.
